Studies on Shoot - tip Culture of M9 , M26 , and MM106 Rootstocks



Abstract
The feasibility of mass propagation of dwarf apple rootstocks through micropropagation technique was studied. Shoot tips of M9, M26, and MM106, sampled on May 23. June 19. July 20. 1980, and May 21, June 12, June 21. July 14, and Sept. 3. 1981, were cultural aseptically on basal Murashige-Skoos¢¥s medium supplemented with various growth regulators. And the results are summarized as follows.
1. Highest per cent callus formation and subsequent favorable growth mere obtained when explants were cultured on a basal medium with NAA 1.0§·/§¤ + kinetin 1.0§·/§¤. BA. when applied singly or in combination, and activated charcoal were also effective.
2. The rate of callus formation decreased as the sampling date was delayed, failing to form the callus is M26 sampled on July 20. But in MM106, callus forming ability was recovered when sampled later (Sept. 3).
3. Varietal differences is shoot differentiation and growth from induced and MM106 shoot tips, callus were significant showing 60%, 40%, and 0 % differentiation in the respective M9, M26, sampled on May 28 and cultured on the medium supplemented with kinetin 2.0§·/§¤.
4. Phloroglucinol(PG) was effective in the differentiation and growth of explants sampled on June 19, but the shoot tips sampled thereafter failed to differentiate.
5. Brown or dark brown callus was induced by auxin, kinetin and BA, and the state of the resulting callus was respectively friable and hard, whereas green on light green hard callus occurred when both auxin and kinetic were added except in MM106. Activated charcoal resulted in the light brown friable callus.
6. The kind of rootstocks and growth regulators affected the fresh and dry weight of callus. NAA increased the weight at higher (MM106) or lower(M26) levels, while stimulating effect of higher kinetin levels was noted, resulting overall callus weight as of the order of M9, M26, and MM106. NAA 1.0§·/§¤ + kinetin 1.0§·/§¤ was most effective in increasing the callus weight in 3 clonal rootstocks tested.
